چکیده

A model for interaction of class A G protein-coupled receptor with the G protein G subunit is proposed using the rhodopsin-transducin (RD/Gt) prototype. The model combines the resolved interactions/distances, essential in the active RD*/Gt system, with the structure of Gt C-terminal peptide bound to RD* while stabilizing it. Assuming the interactions involve conserved parts of the partners, the model specifies the conserved Helix 2 non-polar X-X, Helix 3 DRY and Helix 7/8 NP-YF RD* motifs interacting with the Gt C-terminal peptide, in compliance with the structure of the latter. A concomitant role of Gt and Gt C-termini in stabilizing RD* could possibly be resolved assuming a receptor dimer as requisite for G protein activation.

G-protein Coupled Receptor Dimerization

A growing body of evidence suggests that GPCRs exist and function as dimers or higher oligomers. The evidence for GPCR dimerization comes from biochemical, biophysical and functional studies. In addition, researchers have shown the occurrence of heterodimerization between different members of the GPCR family. Two receptors can interact with each other to make a dimer through their extracellular...

High glucose condition down-regulates the inhibitory G-protein subunit, Gαi, in pheochromocytoma PC12 cells

Introduction: G-proteins have an important role in the cell signaling of numerous receptors. The situation of G-proteins in health and disease and their critical role in the development of diabetic side effects is an interested scientific field. Here, the changes in the expression of G-protein subunits (Gαi, Gαs and Gβ) were evaluated in hyperglycemic situation of PC12 cells as...

P-164: Human Leukocyte Antigen Class Ib and Pregnancy Success

Background: During pregnancy, the maternal immune system is in close contact with cells and tissue from the semiallogenic fetus .The Human Leucocyte Antigen (HLA) class Ib molecules, HLA-E, -F and -G, are expressed at the materno-fetal interface. Because of the apparent immunoregulatory functions of these proteins, they may be involved in successful acceptance of the semi-allogenic fetus during...

Large-scale functional expression of visual pigments: towards high-resolution structural and mechanistic insight.

Introduction In response to endogenous and exogenous stimuli, multicellular organisms develop highly co-ordinated responses. An essential element in this communication system involves G-protein-coupled receptors (GPCRs). This ubiquitous class of membrane proteins arose early during evolution and has developed into one of the largest protein superfamilies known in nature. GPCRs have been identif...
